Unhappy electrophysiology slices or isolated cells?

Hi, I was wondering if anyone could help me figure this out...
i am voltage-clamp recording from isolated neuroblastoma cells to see the effects of transfection of a beta subunit on the voltage-dependent proerties of the channel. I wanted to know what are the major limitations if i wanted to take these recordings from brain slices instead of isolated cells?
any help would be greatly appreciated! Thanks
